The Laboratory of biological electron microscopy and structural biology

(dr. fei sun’s lab)

The Laboratory of Biological Electron Microscopy and Structural Biology(LBEMSB) is an individual research group with the principal investigator, Dr. Fei Sun. It belongs to the National Laboratory of Biomacromolecules, Institute of Biophysics, Chinese Academy of Sciences (CAS). Besides, Dr. Sun also takes the chief responsibility of the Center for Biological Imaging (CBI), Core Facility for Protein Sciences, CAS, which is well equipped. Our research group is one of the major users of CBI.

Our research will combine X-ray crystallography and cryo-electron microscopy approaches as well as other biophysical and biochemical tools to study the molecular architecture of biological systems from nano-scale to meso-scale. In the next five years, we will focus on (i) Molecular mechanism of mitochondrial and bio-membrane dynamics, (ii) Structure and function of supra macromolecular complexes, and (iii) Methodology development on the biological imaging.

In collaboration with  Center for Biological Imaging, IBP, CAS, we developed a new workflow (HOPE-cryoCLEM) of cryo correlative-fluorescence and electron microscopy (cryo-CLEM), based on a high vacuum optical platform and a conventional cryo-EM holder. This workflow can increase the successful rate of cryo-CLEM and is suitable to adopt for super resolution imaging in the future. This design of HOPE has been patented and our work has been published by Journal of Structural Biology recently.
